For "I hope," use 希望 (xīwàng) plus a full clause: 希望你住得舒服 (xīwàng nǐ zhù de shūfu). Notice that Chinese does not add an article or change the verb for a plural person: 祝你们 (zhù nǐmen) works the same way.
When to use it
Use 祝你玩得开心 when someone is about to do something enjoyable: a sightseeing day, a concert, a party. Use 祝你旅途愉快 when the focus is the journey itself, such as before a flight or a long train ride. For a hotel stay, 祝你在这里过得愉快 or 希望你住得舒服 sounds warmer and more specific. 慢慢来，不着急 is different: it tells a person there is no need to hurry, so do not use it as a translation of "enjoy your stay." If you are the guest, you can reply 谢谢，你也是 (xièxie, nǐ yě shì), "Thanks, you too."

祝 (zhù) wishes are common in everyday Mandarin and sound warm without being overly formal. With strangers or staff, 祝你... is fine; with close friends, people often shorten it to 玩得开心！ (wán de kāixīn). 愉快 (yúkuài) is a little more formal than 开心 (kāixīn), so it suits journeys, work trips and written messages. Politeness in Chinese often comes from tone and context rather than adding many words.
One thing to watch for
Translating "enjoy your stay" as 慢慢来 (mànmàn lái) because English "enjoy" and "slowly" seem related.
Try this instead: 慢慢来 means "take your time, no rush." For an enjoyable stay, say 祝你在这里过得愉快 (zhù nǐ zài zhèlǐ guò de yúkuài) or 祝你玩得开心 (zhù nǐ wán de kāixīn). Use 慢慢来 only when someone is rushing, for example: 慢慢来，不着急 (mànmàn lái, bù zháojí).
