In Japanese hotels, requests are usually made politely and indirectly. Using お願いします (onegaishimasu) is standard and safe. Staff may use humble keigo like かしこまりました (kashikomarimashita) and お届けします (otodoke shimasu); you do not need to reply in keigo. A simple ありがとうございます (arigatō gozaimasu) is enough. Tipping is not customary in Japan, so do not offer money for delivery.
One thing to watch for
Using the wrong counter, such as タオルを二つ (taoru o futatsu) for two towels.
Try this instead: Towels are flat, so use the counter 枚 (mai): タオルを二枚 (taoru o nimai). 二つ (futatsu) is for generic objects and sounds less natural for towels. For toothbrushes, use 本 (hon): 歯ブラシを二本 (haburashi o nihon).
