The core pattern is: Kann ich meinen Aufenthalt um + number + Nacht/Nächte + verlängern? For one night use the singular eine Nacht; for two or more use the plural Nächte, for example Kann ich meinen Aufenthalt um zwei Nächte verlängern? The verb verlängern means 'to extend' and goes at the end of the sentence in this question. If you prefer a softer tone, replace Kann ich with Könnte ich: Könnte ich meinen Aufenthalt um eine Nacht verlängern?
When to use it
Use these questions at reception when you are already checked in, not when making a first booking. For a first booking you would say Ich möchte ein Zimmer buchen. To check availability first, ask Haben Sie noch ein Zimmer frei? If the answer is Ja, ein Zimmer ist noch frei, you can then ask to extend. If the answer is Leider ist alles ausgebucht, the hotel has no free room. In Austria and southern Germany you may also hear the word Nacht in the same way; the standard forms above are understood everywhere.

German hotel staff usually appreciate a direct but polite question. Using Könnte ich instead of Kann ich sounds slightly softer, and adding bitte at the end is common: Könnte ich meinen Aufenthalt um eine Nacht verlängern, bitte? In formal situations, address staff with Sie, not du. In Austria, 'Grüß Gott' is a common greeting; in Switzerland, 'Grüezi'. These greetings replace Guten Tag but do not change the grammar of your request.
One thing to watch for
Saying Ich will meinen Aufenthalt verlängern, which sounds demanding, or using buchen for an extension.
Try this instead: Use Ich möchte or Könnte ich for politeness, and use verlängern for an extension: Könnte ich meinen Aufenthalt um eine Nacht verlängern? Reserve buchen for a new booking: Ich möchte ein Zimmer buchen.
